Saliva Hormone Opendx Supreme
At-home saliva collection | Nationwide shipping
Overview
The Saliva Hormone Opendx Supreme test provides a precise, non-invasive assessment of 11 key hormones—measured across multiple time points—using liquid chromatography-tandem mass spectrometry (LC/MS-MS). This highly accurate method detects even low concentrations of hormones, making it superior to immunoassay testing for both men and women.
It includes a four-point cortisol profile to evaluate adrenal function, in addition to essential sex hormones and melatonin, offering deep insight into the balance between stress, sleep, metabolism, and reproductive health.

Hormones Measured & Clinical Relevance
Sex & Reproductive Hormones
- Testosterone
- Primary male sex hormone (also important in women). Influences libido, energy, muscle mass, and mood. Low levels can cause fatigue, depression, and decreased vitality.
- DHT (Dihydrotestosterone)
- A potent derivative of testosterone. Elevated levels are linked to acne, hair loss (androgenic alopecia), and prostate enlargement.
- Androstenedione
- Precursor to both estrogens and androgens. Helps detect adrenal or gonadal dysfunction. Elevated in PCOS or adrenal hyperplasia.
- Progesterone
- Produced after ovulation. Supports menstrual cycle balance, fertility, mood, and sleep. Low levels can lead to estrogen dominance, PMS, or miscarriage risk.
- 17-Hydroxyprogesterone
- A key adrenal steroid precursor. Elevated in congenital adrenal hyperplasia (CAH) or stress; useful in differentiating adrenal from ovarian hormone issues.
- Estrone (E1)
- Weak estrogen; becomes the dominant form after menopause. Excess may indicate estrogen dominance or poor detoxification.
- Estradiol (E2)
- The most active estrogen during reproductive years. Essential for menstrual health, bone density, mood stability, and skin elasticity.
- Estriol (E3)
- Protective and mild estrogen. Elevated in pregnancy; low levels may indicate estrogen deficiency.
Adrenal Function – Diurnal Cortisol (4 points)
Assesses cortisol at four strategic times to evaluate adrenal gland output and circadian rhythm.
Ideal for identifying adrenal fatigue, burnout, or chronic stress responses.
- Cortisol – Morning (30 minutes after waking)
- Reflects the Cortisol Awakening Response (CAR). Should be the highest point of the day. Blunted response suggests adrenal fatigue or HPA-axis dysfunction.
- Cortisol – Noon
- Measures midday adrenal activity. Helps determine how well your energy and stress hormones are maintained through the day.
- Cortisol – Evening
- Should begin to decline. Elevated results suggest inability to wind down, commonly seen with chronic stress, anxiety, or insomnia.
- Cortisol – Night
- Final cortisol level before sleep. Elevated levels are associated with poor sleep quality, nighttime waking, and weight gain.
Melatonin (2 points)
- Melatonin – Morning
- Should be low in the morning. Elevated morning melatonin may signal circadian rhythm disruption or pineal gland dysfunction.
- Melatonin – Night
- Reflects nighttime production. Low levels are linked to insomnia, difficulty falling asleep, and mood disorders.
